Cotton powdery mildew is caused by the fungus Oidiopsis gossypii (syn. Leveillula gossypii), which belongs to the order Erysiphales. Unlike many other powdery mildew fungi, it possesses an internal (endophytic) mycelium, growing inside the plant tissues.

The fungus infects the host through stomata. After colonization, the conidiophores emerge in fascicles (bunches) through the stomatal openings, which is a unique characteristic for identification.
The life cycle involves the spread of asexual conidia by air currents. The pathogen requires specific environmental conditions to thrive, typically involving a combination of high humidity and moderate temperatures.
In the field, the fungus overwinters in crop debris or on alternative weed hosts belonging to the Malvaceae family. This reservoir maintenance is a critical factor in the onset of epidemics.
Accurate field identification requires a hand lens to distinguish the fasciculate conidiophore growth from the more common superficial white powdery mildew fungi that cover the surface entirely.
